4) Sam getting captured by the Yellow-Eyed Demon

The Yellow-Eyed Demon, Azazel, captures Sam along with other special children and forces them to fight each other in season 2. However, Sam gets killed by Jake. This was a pivotal point in the show as we see Dean making a deal with the Crossroads demon to get Sam back. This was a difficult situation for both of them and showed the brotherly bond they had.
